复制叉式复制大肠杆菌等原核生物的环状染色体dna复制时,首先在dna的复制起点上解螺旋。dnab蛋白结合在复制起点处两个解旋了的单链上,分别形成两个前导链(leading strand)的引物(primer),当前导链朝正反两个方向同时延伸时,dnab蛋白朝延伸方向作逆向移动,陆续形成后随链(laggingstrand)的引物。这是dna双向复制的方式。在复制启动时,尚未解开螺旋的亲代双链dna同新合成的两条子代双链dna的交界处,就称为复制叉(replication fork)。两个靠得很近的复制叉之间形成的空间称为“复制泡”(replication bubblo)。真核生物线状双链dna分子复制时也是先形成复制叉,只是复制起点不止一个。复制开始时,dna解旋酶把dna双链分子解开成两股单链,形成了y形的复制叉。复制叉是不对称的,即一条子链是连续合成的,这是前导链,其合成稍稍领先于另一条不连续合成的后随链。由于dna子链的合成是从5,端到3’端方向进行的,所以前导链只需在复制起点上有一个引物,一旦形成复制叉后,dna聚合酶就可沿着亲链模板不断地加上新的核苷酸,从而合成一条新的连续的子链。同样道理,由于dna子链的合成是从5,朝3,方向进行,所以一定要等前导链开始合成而将后随链的合成模板暴露出来以后,后随链的合成才得以进行。此时,先由dna引物酶合成与后随链亲链的碱基互补的、长约10个核苷酸的rna引物,在dna聚合酶作用下沿着后随链模板合成dna,一直延伸到前一个dna片段5’端上rna引物为止。这样合成的是一系列不连续的长约200个核苷酸的片段,这被称为冈崎片段(okazaki fragment)。专一识别dna/rna双链体中的rna链的dna修复酶,切除rna引物,代之以由dna聚合酶合成的dna小片段。最后由dna连接酶把冈崎片段连接成一条连续的dna单链。
