激光散射法测定精子运动特征激光散射法测定精子运动特征诊法。即 用激光散射光谱测定精子运动以分析其特征的一种方法。尤适于对精子运动速度分布尚不清 楚的研究中。因其可对精子运动速度进行即时的直接测定而优于它法之先摄取精子活动, 事后对照片或录像进行分析。本法用氦-氖气激光发生器发射的激光束,通过中性不透明滤 片衰减到15m之低能激光束,经聚焦后通过内置04ml受试样本的活动扁玻璃比色杯( 光通路01cm),精子活动使激光发生散射而得到相应之光谱。该光谱之测定可以快速 傅立 叶转换数字光谱分析仪按照其能谱强弱分析,亦可由光强度衰减时间束分析。精液样本液化 后,取1ml样本加入4ml含04%牛血清白蛋白和pH为74的Hepes缓冲液的BWW培养液,振 荡后 用350~600g离心8分钟,倾弃大部上清液留约1ml,加4mlBWW液混匀后离心6分钟,同法经 第三次洗涤(避免假象及计数误差)离心3分钟。50分钟后即可测定精子活动。90分钟后可再 次测定,以提供对精子活力的估计。正常时,精子运动速度分布接近泊松分布。示性速度 Vc(大多数精子将按此速泳动)、平均速度V-和均方根速度V-1的相互关系 可按Vc∶V-∶V1=1∶2∶24的比例来表达。本法操作简便、数据客观、对 精子无 伤害,况其敏感度高,足以确定化合物剂量反应关系和进行不同化合物的药效比较,能提供 化合物对精子活动影响的敏感参数。
